The long term goalof Dr. Zhong is to explore molecular mechanism of cancer development by determining deregulation of RNA Pol III-dependent gene transcription and to establish the role of chromatin remodeling during cell transformation and tumor development by identifying specific signaling pathway which may involve Pol III gene transcription and histone modification and establishing the epigenetic regulation of transcription machinery of Pol III genes. His current projects focus on the role of alcohol-induced deregulation of Pol III genes in alcohol-associated tumor development; how alcohol-induced histone H3 phosphorylation (H3ph), such as H3S10ph and H3S28ph, mediate transcription machinery of Pol III genes; whether JNK and MSK1 mediate alcohol-induced transcription of Brf1 and Pol III genes; whether blocking the signals inhibit alcohol (or other agents) induces (or represses) cell transformation and tumor formation. His groupis studying how alcohol promotes development of human breast cancer and whether Runx2 and ERα affect transcription factor-Brf1, which specifically regulates Pol III gene transcription. He has published dozens of papers on signal transduction, epigenetic regulation, histone modification and Pol III gene transcription in the peer-review journals. Dr. Zhong expects that these incomes from our studies will elevate our understanding of molecular mechanism of alcohol-associated cancer development and provide potential therapy for cancer patients.
